King oyster mushroom production has three mainstream inoculation methods: solid spawn, wood-dowel (twig) spawn and liquid spawn. Each method has strict environmental requirements and standard operating procedures to lower contamination risk and boost mycelium germination rate.
Start inoculation only after the substrate temperature drops below 25°C. In commercial workshops, sterilization equipment runs 4–5 hours in advance, and the air purification system is turned on 1 hour before work to maintain positive pressure inside the inoculation room. Workers need full sterile protective clothing, and hands & tools are disinfected with 75% alcohol (tools also need flame sterilization). Remove the top aged 1–2 cm mycelium layer from spawn before use. Fill broken spawn into the pre-drilled holes of substrate bags all the way to the bottom. The standard dosage fills the holes and forms an even 0.3 cm spawn layer on the substrate surface, supporting fast and uniform mycelium run.
Wood-dowel spawn should be used 10 days after full colonization for post-maturation. This step ensures mycelium fully penetrates the wood and delivers higher survival rate. Using it immediately after colonization easily causes non-germinating dead spawn. Wipe the spawn surface with alcohol, scrape off aged hard mycelium, take out the dowel spawn and insert it into pre-drilled holes of sterilized substrate bags, then seal the lids.
The cleanroom needs central air filtration, refrigeration and ozone disinfection systems, separated into primary cooling, rapid cooling, pre-inoculation, inoculation and spawn handling areas. An airlock buffer zone and disinfection sink are required at the entrance. The inoculation room adopts Class 100 (ISO 5) filtration and automated conveyor lines; the overall workshop environment keeps Class 10,000 (ISO 7) cleanliness.
Cool cultivation bottles or bags below 25°C first. Run all filtration and disinfection systems following standard SOP, and enforce strict personnel sanitation rules. Cooled containers are sent by conveyor under high-efficiency filters. An automatic liquid inoculator sprays fermented liquid spawn at 0.20 MPa tank pressure, with 25–30 ml dosage per bottle/bag. After inoculation, crates are conveyed out and transferred to the incubation room. For detailed operation, you may refer to enoki mushroom liquid spawn inoculation specifications.
