Key Points of Liquid Spawn Production in Fermentation Tanks

(1) Process Flow

Fermentation tank cleaning and inspection → Empty tank sterilization → Liquid medium preparation → Tank filling → In‑situ medium sterilization → Inoculation → Fermentation and cultivation → Sampling and testing → Fermentation endpoint judgment → Liquid spawn harvest

(2) Medium Formula

Potato 100 g, brown sugar 15 g, glucose 10 g, wheat bran 40 g, peptone 2.0 g, potassium dihydrogen phosphate 2.0 g, magnesium sulfate 1.0 g, Vitamin B₁ (1 tablet), antifoaming agent 0.3 mL, pH 6.8‑7.0, water 1000 mL.

(3) Filling and Sterilization

Prepare medium according to the formula and fill into the fermentation tank. (Example: 50 L medium for a 70 L‑rated tank). Perform thermal sterilization at 121 °C for 1 hour.

(4) Inoculation

Cool the sterilized medium down to below 25 °C. Carry out tank inoculation under strict aseptic conditions. The inoculation volume of primary seed is 1000‑1500 mL per tank.

(5) Cultivation Parameters

Temperature: 25 °C; pH: 6.5; cultivation time: 72‑90 h; tank pressure: 0.02‑0.04 MPa; aeration‑volume ratio: 1:0.8 (v/v/min)
