Production of King Oyster Mushroom Mother Spawn, Primary Spawn, and Cultivation Spawn

(I) Mother Spawn Production

1. Commonly Used Slant Culture Medium Formulas for Mother Spawn

(1) Potato Glucose Agar (PDA)

Potatoes (peeled) 200g, glucose 20g, agar 18-20g, pH 6.5-7.0, water 1000ml.

(2) Potato Composite Culture Medium

Potatoes (peeled) 200g, glucose 20g, agar 18-20g, potassium dihydrogen phosphate 3g, magnesium sulfate 1.5g, 

pH 6.5-7.0, water 1000ml. (3) Rejuvenating Culture Medium: Potato (peeled) 200g, glucose 20g, peptone 5g, agar 

18-20g, potassium dihydrogen phosphate 3g, magnesium sulfate 1.5g, pH 6.5-7.0, water 1000ml.

2. Preparation of Mother Culture Medium: Prepare according to conventional methods.

3. Propagation and Culture of Mother Culture: Inoculate a healthy, vigorous, and bacteria-free mother culture onto

a sterilized blank agar slant medium in a sterilized aseptic workbench. Then, incubate at 25℃ for 7 days to obtain

the mother culture used in production.

2. Selection of Culture Bottles and Bags: The original culture (cultivation culture) can be prepared using standard 

750ml culture bottles and polypropylene bags measuring 17cm long, 33cm wide, and 0.004cm thick. Considering 

ease of operation and reducing infection rate, the author recommends using culture bottles. 3. Production Process

of Primary and Cultivated Strains

Cultivation → Obtaining Primary Strains

4. Propagation and Cultivation of Primary and Cultivated Strains

Superior mother cultures are inoculated into sterilized primary culture medium in a sterilized aseptic operating table. One test tube of mother culture can inoculate 4-5 bottles of primary culture. After inoculation, immediately transfer to a 25℃ incubator or culture room for cultivation. The bottles will be fully colonized in 30 days. Once the primary culture has grown well, it can be used to propagate the cultivated culture. The culture medium formula and preparation for the cultivated culture are the same as for the primary culture, but the cultivated culture grows much faster, reaching full colony size in 25-30 days.
