Preparation of Shiitake Mushroom Mother Culture

(1) Formula

① Potato 200g, Glucose 20g, Peptone 5g, Agar 18-20g, Potassium dihydrogen phosphate 3g, Magnesium sulfate 1.5g, pH 6.5-7.0, Water 1000mL.

② Potato 200g, Glucose 20g, Wheat bran 50g, Agar 18-20g, Potassium dihydrogen phosphate 3g, Magnesium sulfate 1.5g, pH 6.5-7.0, Water 1000mL.

(2) Inoculation and Culture: Prepare, sterilize, and inoculate according to standard procedures, then culture in an incubator at 25℃ in the dark.

(3) Labeling and Culture: Before removing the test tubes from the inoculation chamber, label each tube on the upper front with the strain number and inoculation date. Then place the test tubes in a culture incubator (room) at 23-25℃ for 8-10 days until the slant is fully covered with mycelium.

(4) Strain Preservation: Select uncontaminated culture slants, wrap them in paper, and store them in a refrigerator at 4℃.
